Reducing risk in HPLC / LC-MS therapeutic drug monitoring and treatment: Why water purity is key

Understanding the role of ultrapure water for accurate therapeutic drug monitoring using HPLC-based methods

This white paper will help you to understand just how important ultrapure water is for HPLC and LC-MS/MS by looking at some key applications of HPLC in clinical diagnostics. It examines the challenges that are frequently directly related to the quality of the water used. Applications we look at include (i) ISD monitoring and treatment, (ii) monitoring of anti-psychotic drugs, (iii) drugs testing in forensic toxicology, and (iv) measuring the circulating levels of antimicrobials in severe infection.

What’s inside?

Water is critical to the mobile phase in HPLC and LC-MS/MS. We discuss why water purity is key to applications of HPLC and mass spectrometry in clinical diagnostics.

  • LC-MS/MS for therapeutic drug monitoring (TDM) and treatment. Ultrapure water is needed to deal with threshold quantities of immunosuppressant drugs (ISDs) where there is a low minimum toxic concentration.
  • uHPLC-MS/MS method for TDM of anti-psychotic drugs. Ultrapure water was used in developing a sensitive uHPLCMS/ MS method to co-quantify 7 typical antipsychotic drugs.
  • Sample preparation and processing in forensic toxicology. ELGA water systems are used in LC-MS/MS for hair analysis to detect and quantify different drugs.
  • Measuring antibiotics in severe infections. Ultrapure water is needed for accurate measurement of serum concentrations of antimicrobials with narrow therapeutic index drugs.

The purity of the water used in sample preparation and HPLC ahead of MS is critical, in order to drive accurate and successful patient and medico-legal outcomes.
